Genetik hepatologischer und onkologischer Erkrankungen, Systembiologie, Bioinformatik.
 
Systembiologischen und bioinformatischen Ansätzen in der biomedizinischen Grundlagenforschung kommen in den nächsten 20 Jahren eine wegweisende Rolle bei der Etablierung einer Individualisierung der medizinischen Versorgung zu. Mit einer solchen Individualisierung der Therapie sind eine Reihe von Erwartungen verknüpft: Eine Erhöhung der Genauigkeit von Krankheitsdiagnosen und –prognosen, eine treffsichere Früherkennung von Risikopersonen und Krankheiten, eine gezielte Auswahl wirksamer und nebenwirkungsarmer Therapieoptionen und eine bessere Verlaufskontrolle von Krankheiten.

Wir beschäftigen uns daher insbesondere mit der Identifizierung von prognostisch und therapeutisch relevanten Subgruppen von Patienten mit Leberfibrose und HCC. Entsprechende Patientengruppen werden durch genetische Signaturen auf der Ebene der Genexpression, dem Transkriptom, erarbeitet und definiert. Darüber hinaus hat unsere Arbeitsgruppe in den vergangenen Jahren umfangreiche bioinformatische Ressourcen im Bereich der Hepatologie geschaffen und diese öffentlich zugänglich unter http://www.medicalgenomics.org ins Internet gestellt.
Parallel haben wir in den vergangenen Jahren ein molekularbiologischen Labor aufgebaut, indem derzeit mehrere genveränderte Mausmodelle auf Ihre Entwicklung einer Leberfibrose und HCC untersucht werden. Dabei sind alle Techniken der Generierung genveränderter Mäuse von der Klonierung eines Targetingvektors, ES-Zellkultur und ES-Zell-Injektion im Labor etabliert.

Die Synergie aus molekularbiologischer und bioinformatischer Modellierung ermöglicht eine unmittelbare molekularbiologische Validierung bioinformatisch generierter Hypothesen aber auch eine systembiologische Betrachtung der Leberfibrose und HCC-Entstehung.
