Wir sind an der Entschlüsselung genregulatorischer Prozesse interessiert, wie diese durch exogene und endogene Einflüsse moduliert werden und dies komplexe pathophysiologische Zusammenhänge - wie etwa die rätselhafte Liaison von Blutgerinnung und Tumorbiologie - zu erklären hilft. Von besonderem Interesse sind darüber hinaus die wechselseitigen Interaktionen von vaskulärer Biologie, Inflammation und Immunität.

Wir verfolgen hierbei einen translationalen Forschungsansatz, der Elemente der Grundlagen-forschung (Biochemie, Molekular- und Zellbiologie), die Modellierung von Krankheiten in Tieren und schließlich die Analyse von Patienten vereint. Durch die Verwendung neuer diagnostischer Tools wie die präklinische (molekulare) optische Bildgebung versuchen wir diese Prozesse nicht-invasiv in lebenden Organismen zu visualisieren. Unser Ziel ist es, dieses Wissen schließlich in neue therapeutische und diagnostische Konzepte einfließen zu lassen. Beispiele sind RNA Therapeutika zur Regulation des hämostatischen Systems (Nourse & Danckwardt. Pharmacol Ther 2021, Nourse et al. J Thromb Haemost 2018) sowie RNA-Signaturen als hochpotente neuartige Biomarker in der Onkologie und bei weiteren Erkrankungen (Ogorodnikov et al. Nature Comm. 2018; Marini et al. Nucleic Acids Res. 2021).

Forschungsschwerpunkte

  • Posttranskriptionelle Regulation in der vaskulären Medizin und darüber hinaus
  • Rolle der Blutgerinnung in der Tumorbiologie und darüber hinaus
  • Bedeutung der RNA Regulation in der Entwicklung und bei Krankheiten (non coding RNAs, miRNAs, RNA binding proteins)

Wichtige Elemente sind hierbei genetische Mausmodelle sowie non-invasive preclinical optical imaging. Zudem entwickeln und nutzen wir next generation sequenicing (NGS)-basierte diagnostische Techniken (sCLIP, TRENDseq, miTRAP), um Mechanismen der RNA Regulation zu entschlüsseln und in großem Maßstab funktionelle RNAi Analysen durchzuführen. Wir generieren und veröffentlichen Ressourcen, die über die Grenzen unterschiedlicher Disziplinen hinweg translationale Forschungsaktivitäten fördern und stärken (z.B. TREND-DB: explore interactively the dynamic landscape of transcriptome 3'end diversification).

It is a great privilege! We are now part of the DFG Priority programme 1935 "Deciphering the mRNP code: RNA bound determinants of post-transcriptional gene regulation". In this context, we will address the role of transcriptome 3'end mRNP dynamics on neuronal development.
