Aktuelle Forschung

Infektionskrankheiten stellen nach wie vor ein zentrales globales Gesundheitsproblem dar. In Europa sind Infektionen der unteren Atemwege, die zum Beispiel durch Streptococcus pneumoniae verursacht werden, eine der Hauptursachen für Morbidität und Mortalität, obwohl Impfstoffe gegen diesen Erreger zur Verfügung stehen. Darüber hinaus stellt die Zunahme von Infektionen mit multiresistenten Bakterien, z.B. Mycobacterium tuberculosis, eine ständige Bedrohung für unsere Gesellschaft dar. Unter dem Dach der Abteilung für Medizinische Mikrobiologie und Hygiene untersuchen Forscher um Univ.-Prof. Dr. Tim Sparwasser mehrere zentrale Aspekte der Infektionsbiologie. Dazu gehört ein besseres Verständnis der Wirt-Pathogen-Interaktionen und der Mechanismen, die die infektionsbedingte Immunantwort aktivieren und regulieren. Darüber hinaus erforschen die Wissenschaftler den genetischen Hintergrund und die molekularen Prozesse der infektionsassoziierten Immunantwort und sie suchen nach neuen Wegen, die Immunantwort zu modulieren. Das Ziel der Arbeiten ist es schließlich, die neuen Erkenntnisse der Grundlagenforschung in die klinische Anwendung zu übertragen.

 

Fortschritte in der modernen Medizin, insbesondere auf dem Gebiet der Transplantation und der Intensivmedizin, haben die Lebenserwartung in der westlichen Welt stark erhöht, führen aber auch zu einem Wiederauftreten opportunistischer (z.B. CMV und Candida) und im Krankenhaus erworbener (z.B. Clostridium difficile) Infektionen, die unser medizinisches System vor ernste Probleme stellen. Weitere Herausforderungen der alternden europäischen Gesellschaft sind Autoimmunität und Krebs. Die gemeinsame Ursache dieser Hauptherausforderungen liegt in eine unzureichenden T-Zell Immunantwort.

 

Impfungen gelten als eine der größten Erfolgsgeschichten in der Medizin und können langfristige Nachhaltigkeit und Effizienz garantieren. Die Identifizierung verbesserter Adjuvantien, insbesondere für die T-Zell-Immunität, würde eines der größten klinischen Probleme lösen. Neuartige Impfstoffe, insbesondere gegen weitgehend arzneimittelresistente Bakterien wie Mykobakterien, stellen nach wie vor eine große Herausforderung dar. Ein neuer Ansatz, bei dem nicht der Erreger selbst im Mittelpunkt steht, ist das Konzept der "Immunmodulation". Die Identifizierung und Charakterisierung von Stoffwechselprodukten der Mikrobiota zeigt, dass diese einen direkten Einfluss auf die Immunreaktion des Wirts haben, neue therapeutische Möglichkeiten zur Verkürzung der Antibiotikatherapie aufzeigen oder zur Verringerung der Immunpathologie bei Infektions- oder Entzündungskrankheiten beitragen.
