TRP X26 - Coagulation Proteinases in human monocyte differentiation

Project Summary

Monocytes play a key role at the interface between inflammation and coagulation by forming coagulation factors but also reacting to them via protease-activated receptors (PARs). However, with a few exceptions, the influence of most factors or their complexes on defined monocyte subpopulations and their differentiation into macrophages and dendritic cells has not been investigated. Research in this area focused on TF and FVIII mediated effects on differentiated macrophages and dendritic cells. However, the contribution of single coagulation proteases to monocyte differentiation into phagocytes, especially tolerogenic dendritic cells is not addressed so far. The aim of this project is therefore to decipher the effect of coagulation proteases in the differentiation of human monocytes to macrophages and dendritic cells (DC) using recombinant factors and a phased stimulation strategy. The data obtained will provide starting points for further translational investigations on (coagulation factor-deficient and thrombotic) patients.
