TRP X29 - The role of B lymphocytes in venous thrombosis

Project Summary

Recent studies have shown that B lymphocytes play an important role in the development of atherosclerosis and insulin resistance. In contrast, the role of B cells in venous thrombosis is completely unknown. Our preliminary results suggest that B cells may be important regulators of hemostasis. In the current project, we aim to test this hypothesis in a mouse model of venous thrombosis (partial ligation of the inferior vena cava). With adoptive transfer of B-1 and B-2 cells, we will also investigate of the role of B cell subsets in venous thrombosis. The potential role of neutrophilia caused by B cell deficiency will be addressed with anti-Ly6G antibody-induced neutrophil depletion and with genetic disruption of peptidyl arginine deiminase 4 (PAD4), respectively.

The role of B cells in human deep vein thrombosis (DVT) is also unknown. In a pilot study, we will analyze B cell signature, antibody profile and B cell-related cytokines in the blood of DVT patients. In combination with changes in the whole blood expression of B cell-related genes, we may obtain the first information on the role of B cells in human DVT.
