Dozent | Prof Dr Markus Glatzel Abstract: The cellular prion protein (PrPC) is a glycosylphosphatidylinositol (GPI)-anchored protein with two variably occupied N-glycosylation sites, expressed mainly in neurons. Its pathological isoform (PrPSc) accumulates in prion diseases and represents an essential component of the transmissible entity, the “prion”. Besides its key role in prion-diseases, PrPC plays an important role in Alzheimer disease. Here it was recently identified as a receptor for oligomeric forms or beta-amyloid, the neurotoxic species responsible for initiation and progression of Alzheimer disease (AD). Recent data concerning proteolytic processing of PrPC at plasma membranes and it implications for pathophysiology of prion disease and AD will be presented. |
