Advanced Spectral Flow Cytometry Aurora Course

The Advanced Spectral Flow Cytometry Aurora Course is a three-day training program for participants with prior experience in flow cytometry. The course combines theoretical instruction with practical demonstration and training for the Cytek(R) Aurora platform. Participants learn the principles of spectral detection and how complete fluorescence signatures are used to distinguish multiple fluorochromes. A major focus is the rational design of high-dimensional panels based on biological questions, marker co-expression, antigen density, fluorochrome brightness, spectral similarity, and spread. The course also covers antibody titration, blocking reagents, panel validation, and the selection of suitable controls. Participants are introduced to spectral unmixing, reference-control quality, autofluorescence assessment, and troubleshooting of incorrect signatures. During the practical sessions, they learn standardized instrument operation, experiment setup in SpectroFlo, sample and technical control acquisition, post-acquisition unmixing, and quality control in FlowJo. The course concludes with data management, documentation, and a panel-design assignment that integrates the complete workflow from experimental planning to reliable data interpretation.

Target Audience: TransMed Fellows with prior experience in flow cytometry. Completion of a basic course on flow cytometry from the AG MAYER (or equivalent) is required.
